چکیده

We present a fast, high-throughput method for characterizing the motility of microorganisms in 3D based on standard imaging microscopy. Instead of tracking individual cells, we analyse the spatio-temporal fluctuations of the intensity in the sample from time-lapse images and obtain the intermediate scattering function (ISF) of the system. We demonstrate our method on two different types of microorganisms: bacteria, both smooth swimming (run only) and wild type (run and tumble) Escherichia coli, and the bi-flagellate alga Chlamydomonas reinhardtii. We validate the methodology using computer simulations and particle tracking. From the ISF, we are able to extract (i) for E. coli: the swimming speed distribution, the fraction of motile cells and the diffusivity, and (ii) for C. reinhardtii: the swimming speed distribution, the amplitude and frequency of the oscillatory dynamics. In both cases, the motility parameters are averaged over ∼ 10 cells and obtained in a few minutes.

Differential dynamic microscopy of bacterial motility.

We demonstrate a method for the fast, high-throughput characterization of the dynamics of active particles. Specifically, we measure the swimming speed distribution and motile cell fraction in Escherichia coli suspensions. By averaging over ∼10(4) cells, our method is highly accurate compared to conventional tracking, yielding a routine tool for motility characterization. We find that the diffu...

Stability of the Modified Euler Method for Nonlinear Dynamic Analysis of TLP

Efficiency of numerical methods is an important problem in dynamic nonlinear analyses. It is possible to use of numerical methods such as beta-Newmark in order to investigate the structural response behavior of the dynamic systems under random sea wave loads but because of necessity to analysis the offshore systems for extensive time to fatigue study it is important to use of simple stable meth...

O-12: Tubulin Reversible Acetylation – Driving The Moves and The Moves Behind The Drive

Background Asthenozoospermia accounts for almost 50% of the cases of male infertility. Our study investigating phosphoproteins differentially expressed in asthenozoosperm has identified the phosphoproteins relevant to sperm motility and the signature molecules likely to be altered in asthenozoospermia. The 66 phosphoproteins differentially expressed included four alpha tubulin isoforms which we...
